Despite premature senescence of rhizobia in mate67 nodules (Supplemental Fig. S12), residual nitrogenase activity indicated that some rhizobia differentiate fully into bacteroids in these mutant nodules (Fig. 6). Consistent with this, we observed what appeared to be fully mature bacteroids by TEM of 10 dpi nodule sections (Supplemental Fig. S12; c.f. Vasse et al., 1990). Interestingly, numerous small vacuoles in infected nodule cells of mate67 mutant plants were observed (Supplemental Fig. S12C), resembling the sen1 mutant phenotype of L. japonicus (Hakoyama et al., 2012). SEN1 belongs to the family of VIT and is thought to deliver Fe to symbiosomes. Like mate67, sen1 mutants are defective in SNF (Hakoyama et al., 2012; Suganuma et al., 2003).

To elucidate the in vivo effects of Aβpeptide on the retina, C57BL/6 mice received a single unilateralsubretinal injection of Aβ peptide (Fig. 1). Briefly, 5-month-old C57BL/6mice were anesthetized by an intraperitoneal injection of 300 mg/kgchloral hydrate (Sangon Biotech, Shanghai, China). The pupils weredilated with 0.5% tropicamide and 0.5% phenylephrine (SantenPharmaceutical, Osaka, Japan); 0.5% proparacaine hydrochloride(Alcon, Shanghai, China) was applied for topical anesthesia.Following complete pupillary dilation, the anesthetized animalswere placed under an Nikon SMZ-1 stereoscopic microscope, and thecorneas were carefully punctured through the pars plana with a30-gauge needle to produce a hole sufficiently large enought toinsert a 33-gauge blunt needle (Hamilton Co., Reno, NV, USA). Theblunt needle tip was inserted through the corneal puncture andadvanced into the anterior chamber. A slight resistance to themovement of the needle indicated the penetration of the retina andentrance into the subretinal matrix.
